Pgrac

Pgrac is an artificial promoter consisting of Bacillus subtilis groE promoter, lac operator and gsiB ribosome binding site. This is a strong IPTG-inducible promoter for B. subtilis, which was first appeared on plasmid pNDH33 and later on pHT01 of the company MoBiTec 1. Plasmid pHT01 is supposed to be stable in both B. subtilis and Escherichia coli, and can be used for expression of proteins in these host strains. Using pNDH33/pHT01 proteins could be produced up to 16% of total protein in B. subtilis. Pgrac100 is an improved version of Pgrac, which can produce up to 30% of total cellular proteins in B. subtilis.
